BMS-986408 is a first-in-class dual inhibitor of DGKα and DGKζ, exhibiting IC50 values of 0.0003 μM and 0.002 μM for DGKα and DGKζ lipid kinase activity, respectively. It potentiates T cell–receptor signaling and T-cell activation, enhancing the effectiveness of both PD-1 and CAR T-cell therapy in preclinical models.
