1.Flow cytometric analysis of anti-EpCAM CAR expression. 5e5 of anti-EpCAM CAR-293 cells were stained with 100 μL of 5 μg/mL of AF 488-conjugated human EpCAM Protein (Site-Specific AF 488 Conjugation, ECD, His Tag, Cat. No. 10694-H86H-SD) and negative control protein. Non-transduced 293 cells were used as a control (left) . AF 488 signal was used to evaluate the binding activity (QC tested) . 2.Binding activity of AF 488-conjugated EpCAM protein to PBMC cells. PBMC cells were stained with PE conjugated anti-CD3 antibody and AF 488-conjugated human EpCAM Protein (Site-Specific AF 488 Conjugation, ECD, His Tag, Cat. No. 10694-H86H-SD) and detected by flow cytometry. PE signal was used to evaluate the content of CD3+ T cells in PBMCs. AF 488 signal was used to evaluate the non-specific binding activity to PBMCs (QC tested) . 3.Immobilized anti-EpCAM scFv-hIgG1 Fc at 1 μg/mL (100 μL/well) can bind Recombinant Human EpCAM Protein (ECD, His Tag, Site-Specific AF488 Conjugation) (Cat: 10694-H86H-SD), the EC50 is 54.0 ng/mL (Routinely tested) .
