Description

RNA Mango technology is based on the specific binding between the RNA Mango aptamer and a thiazole orange (TO) bi-functional dye. This interaction is characterized by strong affinity (KD ≈ 3 nM) and a ~1000-fold enhancement of dye fluorescence upon binding to the Mango aptamer (fluorescent enhancement FE ≈ 1,100) . The TO dye also has several advantageous properties, including small size, low toxicity, permeability through plasma and nuclear membranes, short intracellular half-life, and tunable spectral properties through structural modifications. TO1-biotin is the standard dye used for RNA Mango experiments, although other variants are also available. RNA Mango enables advanced RNA tracking, visualization, and pull-down applications. Although RNA-based tools have historically lagged behind DNA and protein technologies, RNA Mango provides a powerful approach for studying RNA function in cells. It was developed by researchers including Dr. Peter Unrau of Simon Fraser University. Transcription reactions are typically performed using T7 RNA polymerase in optimized buffer conditions, with TO1-3PEG-biotin dye and appropriate nucleotide concentrations. Samples may include negative controls, DNA templates for transcription, or positive control RNA. Fluorescence is visualized under blue light, often recorded and analyzed for signal enhancement.
